Description

Pharmacokinetics:

Nepafenac, the active ingredient in Nepafenac Ophthalmic Solution, is a prodrug that undergoes rapid conversion to its active metabolite, amfenac, upon penetration into ocular tissues. Once activated, amfenac inhibits the enzyme cyclooxygenase, thereby blocking the production of inflammatory prostaglandins responsible for ocular inflammation and pain. Nepafenac achieves therapeutic concentrations in the aqueous humor of the eye, exerting its anti-inflammatory effects locally with minimal systemic absorption.

Pharmacodynamics:

Nepafenac Ophthalmic Solution acts by reducing inflammation and pain associated with various ocular conditions, including postoperative inflammation following cataract surgery and relief of pain and discomfort associated with ocular surgery. By inhibiting the synthesis of inflammatory mediators, Nepafenac helps to alleviate symptoms such as redness, swelling, and discomfort, promoting ocular comfort and visual recovery.

Drug Classification:

Nepafenac Ophthalmic Solution belongs to the class of medications known as nonsteroidal anti-inflammatory drugs (NSAIDs). It is classified as a selective cyclooxygenase-2 (COX-2) inhibitor, specifically indicated for the treatment of postoperative ocular inflammation and pain.

Formulations:

AIT Pharmaceuticals Division offers Nepafenac Ophthalmic Solution in the following formulation:

Eye Drops: Each ml contains 1 mg of Nepafenac, formulated as a sterile ophthalmic solution for topical administration. The product is packaged in a convenient dropper bottle with a capacity of 5 ml, ensuring easy and precise dosing.

Indications:

Nepafenac Ophthalmic Solution is indicated for the treatment of postoperative inflammation and pain following cataract surgery. It is also indicated for the relief of pain and discomfort associated with ocular surgery, including refractive surgery procedures such as LASIK.

Usage:

To use Nepafenac Ophthalmic Solution, wash your hands thoroughly and tilt your head back. Gently pull down the lower eyelid to create a small pocket and hold the dropper bottle upside down. Instill one drop of the solution into the affected eye(s) while avoiding contact with the dropper tip to prevent contamination. Repeat the process for the other eye if necessary. Use as directed by your healthcare provider.

Dosage:

The recommended dosage of Nepafenac Ophthalmic Solution is one drop instilled into the affected eye(s) three times daily, beginning one day prior to cataract surgery and continuing for two weeks postoperatively. For other ocular surgeries, follow the dosing regimen prescribed by your healthcare provider.

Side Effects:

Common side effects of Nepafenac Ophthalmic Solution may include temporary stinging or burning sensation upon instillation, eye irritation, blurred vision, and increased sensitivity to light. Rare but serious side effects may include corneal epithelial defects, eye pain, and allergic reactions. If you experience any persistent or severe side effects, discontinue use of the medication and consult your healthcare provider promptly.
