Pharmacokinetics:
Polymyxin B Sulphate, an antibiotic belonging to the polymyxin class, exerts its bactericidal action by disrupting the integrity of bacterial cell membranes. Following ocular administration (E. Drop), polymyxin B sulphate is minimally absorbed systemically and primarily acts locally to eliminate susceptible bacteria. The onset of action is rapid, with therapeutic concentrations achieved in the ocular tissues within minutes of application.
Pharmacodynamics:
Polymyxin B Sulphate is classified as a polypeptide antibiotic and is primarily active against gram-negative bacteria, including Pseudomonas aeruginosa. By binding to the lipid component of the bacterial cell membrane, polymyxin B disrupts membrane integrity, leading to leakage of cellular contents and bacterial cell death. This mechanism of action makes it an effective treatment for bacterial conjunctivitis and other ocular infections caused by susceptible organisms.

Formulations:

- Eye Drops: Each ml contains 10,000 units of polymyxin B sulphate in a sterile ophthalmic solution.
Indications:
Polymyxin B Sulphate is indicated for the treatment of bacterial conjunctivitis, keratitis, and other ocular infections caused by susceptible gram-negative organisms. It may be used as monotherapy or in combination with other antimicrobial agents to achieve optimal therapeutic outcomes.
Usage:
Shake the bottle well before each use. Instill one to two drops of Polymyxin B Sulphate into the affected eye(s) every 3 to 4 hours, or as directed by your healthcare provider. Tilt your head back, pull down the lower eyelid, and place the prescribed number of drops into the eye(s) without touching the dropper tip to any surface. Close your eyes gently and apply light pressure to the inner corner of the eye for 1-2 minutes to prevent drainage and systemic absorption.
Dosage:
The recommended dosage of Polymyxin B Sulphate is one to two drops into the affected eye(s) every 3 to 4 hours. Your healthcare provider may adjust the dosage regimen based on the severity of your infection and your individual response to treatment. Do not exceed the prescribed dosage without consulting your healthcare provider.
Side Effects:
Common side effects of Polymyxin B Sulphate may include transient ocular irritation, burning sensation, stinging upon installation, and blurred vision. Allergic reactions such as itching, redness, and swelling of the eyelids or conjunctiva may occur in rare cases. If you experience any unusual symptoms or side effects, discontinue use of the medication and consult your healthcare provider promptly.
